Influence of growth hormones and culture manipulations on globular embryogenic structure formation in blueberries (Vaccinium spp.).

In plant tissue culture isolated plant parts from the intact and healthy plant are used as explants; which kept on the suitable semi-solid (agar gelled) or liquid nutrient media with or without growth hormone. This nutrient media nurture and work as the conductive elements those surrounds the plant parts. The growth regulators in the culture media helps the explants to dedifferentiate and redifferentiate, which eventually helps in callus formation, elongation and proliferation. However, to maintain the plant cells in the in vitro system the three most essential factors are explant selection, media composition and culture condition and the later is very crucial to ‘turn on’ the inherent trait for regeneration. Sometimes growth hormones act as the missing link in the completely optimized culture media, when it does not give the desired result. A new attempt has been made in order to develop a protocol for somatic embryogenesis in blueberries in semi-solid and liquid media in a bioreactor system. Young leaf explants from three low-bush blueberries (V. angustifolium Ait.) were established in semi-solid blueberry basal medium (BM) containing with different combinations of indole-3-acetic acid (IAA) + zeatin; 2,4-Dichlorophenoxyacetic acid (2,4-D) + zeatin and thidiazuron (TDZ). While highest percentage (77%) of callus formed in BM supplemented with IAA (0.1 ml/L) + zeatin (1.5 ml/L), 99% of globular embryogenic structure (GS) formation was recorded in 2,4-D (0.01 ml/L) + zeatin (1.5 ml/L) after 4 weeks of culture. The proliferation rate of GS was greatest when transferred to the liquid BM fortified with TDZ (1.0 ml/L). Although after 13 weeks of culture, the callus turned brown and died in liquid media; GS increased in number and formed secondary (2°) embryogenic structure by transferring them to the semi-solid BM complemented with TDZ (0.5 ml/L). Shoot regeneration in a very less frequency was noticed from the (2°) GS afterwards. Results obtained from this study manifested the influence of growth hormones and culture condition on the somatic embryogenesis in semi-solid and liquid media.
