Phylogenomics Caudillo-Ruiz, Kiela B. [1], Banniza, Sabine [1]. Species identification of the stemphylium blight pathogen in Saskatchewan. The stemphylium blight (SB) pathogen has been reported from around the world as a species in the genus Stemphylium. In Saskatchewan, SB has been little studied and the species identity has not been confirmed yet as it has only been recognized as a potential disease of lentil for the last decade. An accurate species identification of the SB pathogen is essential for the identification of potentially resistant international germplasm. Using 20 isolates collected from infected pulse species throughout Saskatchewan, conidial dimensions were compared with ext-type specimen of S. botryosum. Sequences of the ITS and glyceraldehyde-3-phosphate dehydrogenase (gpd) genes were also compared with sequence data of various species in published data bases to establish a phylogenetic tree. Analysis of conidial dimensions (60 conidia per isolate) revealed that 14 of the 20 isolates overlapped in size, and all isolates from Saskatchewan were different from the ex-type specimen. Of those with different spore dimensions, three also had ITS, but not gpd, sequences different from Stemphylium species. Based on sequence data of both genes, none of the isolates from Saskatchewan was grouped with the ex-type specimen of S. botryosum, but showed high similarity to several other species of Stemphylium. However, further comparisons with type specimens of other Stemphylium spp., if available, will be required to clarify which species infect lentil in Saskatchewan.
