The CLASP microtubule-associated protein modulates and is also a direct transcriptional target of the brassinosteroid signalling pathway during root apical meristem development. In plants, the root meristem is required to provide new cells for sustaining root growth and is regulated by brassinosteroids (BRs) and microtubules. Here, we describe a mechanism wherein BR controls meristem size through microtubules. We show that BR promotes precocious exit of meristematic cells to elongation by CLASP-mediated cortical microtubule reorientation. BRASSINAZOLE-RESISTANT 1 and 2 (BZR1 and BZR2/BES1), the two key transcription factors in the BR signalling pathway, directly target and repress CLASP expression. BR also leads to altered CLASP distribution. clasp-1 null mutants display defects in cortical microtubule pattern and smaller meristems, reminiscent of BR-treated plants. We also find a facilitation effect of CLASP and microtubules on the recycling of the surface receptor, BRASSINOSTEROID-INSENSITIVE 1 (BRI1) to the plasma membrane. clasp-1 mutants show enhanced vacuolar targeting of BRI1 and an impaired BR response.
