Characterization of MOR1-microtubule interactions by observing microtubule dynamics and organization in mor1-tubulin double mutants. MOR1 (MICROTUBULE ORGANIZATION 1) is a microtubule-associated protein with tubulin polymerase and depolymerase activity. It is essential for the regulation of microtubule dynamics and organization, and thus for the control of directional cell expansion by the cortical microtubule array. Mutations in genes encoding tubulin subunits and MOR1 have been shown to impair directional cell expansion, leading to helical organ twisting and cell swelling. We hypothesized that certain tubulin and mor1 mutants exhibit twisting and swelling phenotypes as a result of disruptions to MOR1-tubulin interactions, and created mor1-tubulin double mutants in order to characterize interactions between specific domains of the MOR1 and tubulin proteins. We crossed tubulin mutants with right-handed twisting growth to mor1 mutants with left-handed twisting growth and cell swelling, observing additive/synergistic phenotypes in certain double mutant combinations, which indicated a potential role for the mutated amino acid residues in MOR1-tubulin interactions. We then observed GFP-labelled microtubules in these double mutants, so as to look for effects on specific aspects of microtubule dynamicity and organization. Log in to add this item to your schedule
