Functional characterization of GmMYB176 interacting protein kinases in soybean.

GmMYB176 is an R1 MYB transcription factor that regulates isoflavonoid biosynthesis in soybean. It interacts with the phospho-protein binding proteins, 14-3-3s. Alteration of phosphorylation sites in GmMYB176 affects its interaction with 14-3-3 and its intracellular localization, demonstrating that phosphorylation plays a critical role in nucleo-cytoplasmic shuttling of GmMYB176. Here, using co-immunoprecipitation, we identified 6 putative soybean protein kinases that interact with GmMYB176. The interaction of each of the candidate kinase with GmMYB176 was validated in planta. The results revealed that GmMYB176 interacts with Gm08PK, Gm14PK and Gm17PPD in the nucleus, whereas Gm17PK in the nucleus and the cytoplasm. Even though the candidate protein kinases showed protein-protein interaction with GmMYB176 in the nucleus, they were localized in different subcellular compartments in the cell. Current effort is focussed into heterologous expression of the candidate kinases for kinase assay to evaluate their ability to phosphorylate GmMYB176. The research outcome will provide insight into the regulation of GmMYB176 and isoflavonoid biosynthesis, and thus may help in metabolic engineering of isoflavonoid biosynthesis in plants.
