Biotic and abiotic stress

Response of jasmonic acid deficient tomato mutant (def-1) to salt stress.

Salinity is a major environmental stress factor limiting plant growth and productivity in agriculture. During salt stress, plants must adjust their growth and development under this conditions through the regulation of plant hormones signaling. Jasmonic acid and its metabolites, collectively known as jasmonates are important plant signalling components that mediate responses to abiotic and biotic stresses; however, its role under salinity stress is not completely uncovered. We investigated the response of a jasmonic acid deficient tomato mutant (def-1) to salt stress in comparison to the response of wild type tomato plants. Both genotypes were grown in a hydroponic system in a greenhouse and treated with 50 and 100 mM NaCl for two weeks. Our results showed that def-1 showed less inhibition in biomass as indicated by shoot and root dry weight, leaf area and the number of leaves per plant during salt stress compared to wild type tomato plants. The membrane integrity in def-1 was less impaired by NaCl than in the wild types plants as indicated by electrolyte leakage. In addition, the effects of salt stress was tested on seeds germination of both def-1 and wild type. Salinity stress did not affect the germination rate of both genotypes. However, the roots length of the wild type plants were strongly inhibited by salt stress compered to def-1. Our results showed that the jasmonic acid deficient mutants were less sensitive to salinity stress. One possible role of jasmonic acid during salt stress is that jasmonic acid could mediate oxidative stress. Therefore, the difference between both genotypes in oxidative damage under salt stress will be our future work.
