Molecular phylogeny of Penstemon section Fasciculus (Plantaginaceae) based on single copy orthologous genes (COSII).

Penstemon Mitch. is the largest plant genus endemic to North America, having ca. 280 species. The genus is widely variable in morphological characters, and much of this diversity is associated with evolutionary adaptations such as specialization for pollinators or ecological niches. Previous phylogenetic analyses for the genus used ITS and cpDNA spacer regions. These analyses provided some insights into the phylogenetic relationships within the genus. However, phylogenetic relationships were not always consistent with current taxonomy, and relationships within strongly supported clades were largely unresolved. In the case of the Mexican species of Penstemon, the sampling in the previous analyses was limited. In particular section Fasciculus, which includes about 50 % of the Mexican species, was represented by very few species. For our current study, we used eight single-copy nuclear gene regions from the COSII data set to resolve the relationships between 15 taxa of Penstemon section Fasciculus. The sampling of this study also included five Mexican species from other Penstemon sections as well as several outgroup species. The preliminary results from these analyses show that the section Fasciculus is polyphyletic. However, the sister relationship of P. bolanius, P. campanulatus and P. roseus from subsection Campanulati are well supported, as well as P. gentianoides, P. hartwegii and P. isophyllus from subsection Fasciculi. Based on these results, we conclude that the current circumscription of subsections needs revision to reflect more closely the evolutionary relationships of species.
