Stem rust is a common disease in wheat caused by Puccinia graminis Pers.:Pers. f. sp. tritici Eriks. & E. Henn). Current wheat varieties registered in Canada have good resistance to North American races of stem rust; however, Ug99 (race TTKSK) and its variants pose considerable threat to wheat crops in Canada. While new resistance genes are needed to combat the threat from Ug99, the Sr2 stem rust resistance gene provides broad-spectrum, durable adult plant resistance and has been used for over 60 years in wheat breeding programs around the world. Emmer wheat was the source of the Sr2 gene and it resides on 3BS in hexaploid wheat. Selection for the Sr2 gene is difficult because the inheritance of the gene is recessive, and the expression of the pseudo black chaff phenotype is heavily influenced by genotype and environment interactions. Additionally, wheat breeders usually select against the pseudo-black chaff phenotype to preserve the aesthetic look of wheat plants. Thus, use of molecular markers would be useful in selecting for Sr2 resistance. Simple sequence repeat markers previously developed for Sr2 have been ambiguous due to the nearly identical sequence of the 3BS region around the gene in hexaploid wheat. In contrast, the KBioscience Competitive Allele-Specific Polymerase chain reaction (KASPar) marker for Sr2 targets a precise region of the 3BS region linked to the Sr2 gene. In this study we investigated the applicability of the KASPar marker corresponding to SNP ID wMAS000005 for detecting Sr2 in Canadian wheat varieties used actively as breeding parents.
